This part was built to characterize the mgtA riboswitch. Putting the mgtA riboswitch under the control of the TetR promoter allowed us to test the regulation capabilities of the mgtA riboswitch alone under different concentrations of magnesium. This part was built as a control for the mgtA system. For the experimental protocol click here. This part works as expected. There is a small dorp with increasing concentration of magensium in the circuit with just magnesium. The drop is not quite as dramatic as the K902017. We believe that this is due to the transcription factor PhoP binding to the mgtA promtoer. Since there is no mgtA promtoer in this circuit we believe that the response is not dramatic enough, i.e there are not enough transcripts produced. However we still see some supression with increasing concentration of magnesium. This is due to magnesium binding to the RNA strand and blocking translation and thereby reducing GFP output.

Figure 1: The blue line indicating the response to magnesium shows a dramatic decrease at approximately 1 mM concentration of magnesium. the system completely shuts off at 1mM magnesium.
